HiCEP technology involves 1) synthesizing cDNA from poly (A)+ RNA, 2) cutting with restriction enzymes, 3) adding an adaptor sequence, 4) amplifying the DNA, 5) high resolution electrophoresis, and 6) measurement of peak position and height of the cDNA fragments (Fig. 2). A special restriction enzyme is used for the cutting step which generates representative cDNA fragments for each piece of poly (A)+ RNA. Next, these cDNA fragments may be evenly amplified by using special adaptors and primers.
